Efecto del ayuno durante dos tiempos de confinamiento y de transporte terrestre sobre algunas variables sanguíneas indicadoras de estrés en novillos

RESUMEN
El objetivo de
este estudio fue determinar el efecto de dos períodos de
ayuno en bovinos, ya sea en confinamiento o con transporte
terrestre, sobre las concentraciones plasmáticas de
cortisol, glucosa,
ß-hidroxibutirato y lactato, actividad plasmática de
CK, además de los valores de
leucocitos y VGA. El estudio comprendió un experimento
realizado en julio del 2001 y otro en noviembre del 2002, en la
provincia de Valdivia, Chile.
Se utilizaron 120 novillos Frisón Negro, todos
del mismo predio, de similar edad (dientes de leche y 2
dientes permanentes) y peso vivo (promedio 450 kg). En cada
experimento se les extrajo una muestra de
sangre por
venopunción coccígea a 60 novillos, se
identificaron individualmente y fueron pesados. Luego fueron
asignados al azar en cuatro grupos de 15
animales cada
uno: un grupo fue
confinado en el predio sin agua ni comida
por tres horas (confinamiento 3 h), al mismo tiempo otro
grupo fue transportado en un camión por el mismo tiempo
(transporte 3 h); los otros 2 grupos fueron sometidos a similares
tratamientos respectivamente por 16 h (confinamiento 16 h y
transporte 16 h). Después del tratamiento se les extrajo a
los novillos una segunda muestra de sangre y fueron
pesados.
El cortisol se determinó por radioinmunoensayo
(RIA), la glicemia se midió con el procedimiento
para glucosa GOD-PAP, sin deproteinización; el lactato se
midió con el test UV
enzimático; el ß-hidroxibutirato con la
técnica que utiliza ß-HBA deshidrogenasa para medir
la reducción de NAD+aNADH; el VGA y leucocitos con un
contador hematológico y la actividad plasmática de
CK se determinó con el método
UV-cinético, a 340nm y 37ºC. Los indicadores de
estrés
fueron revisados para determinar su distribución normal mediante
inspección visual del histograma y con la prueba de
Kolmogorov-Smirnov. Cuando ambos, el histograma y la prueba de
Kolmogorov-Smirnov indicaban que la distribución no era
normal las variables se
normalizaron transformándolas a su logaritmo natural. Se
utilizaron modelos
generalizados de regresión
lineal múltiple para determinar las asociaciones entre
los indicadores de estrés y las variables independientes.
Las variables independientes fueron tratamiento (antes del
confinamiento o transporte, confinamiento en ayuno por 3 h,
transporte en ayuno por 3 h, confinamiento en ayuno por 16 h,
transporte en ayuno por 16 h), y la estación del
año (invierno y primavera).
Los novillos transportados por 3 h y aquellos en
confinamiento por 16 h tuvieron concentracion es de Cortisol
significativamente más altas (P<0.01) que antes del
tratamiento; los otros tratamientos y la estación no
afectaron significativamente (P=0.20) los niveles de Cortisol. El
VGA disminuyó significativamente (P<0.01) en los
novillos confinados por 3 h y aumentó significativamente
(P=0.02; P<0.01) en los novillos transportados por 3 y 16 h,
respectivamente. El promedio de VGA fue mayor (P<0.01) en
invierno que en primavera. El recuento de leucocitos no
mostró un patrón consistente, pero aumentó
(P<0.01) en los novillos transportados por 3 y 16 h. La
estación no influyó sobre el recuento leucocitario.
Las concentraciones de glucosa aumentaron significativamente
(P<0.01) con todos los tratamientos y fueron menores
(P<0.01) en invierno que en primavera. Los novillos que
ayunaron confinados por 3 h tuvieron una disminución
significativa (P=0.02) del lactato sanguíneo respecto al
valor inicial.
Los animales ayunados por 16 h, tanto confinados como sometidos a
transporte, tuvieron concentraciones significativamente menores
(P<0.01) de ß-HBA que antes del tratamiento; el promedio
de ß- HBA fue mayor (P<0.01) en invierno. Los novillos
confinados y los transportados por 3 h mostraron actividad
plasmática de CK significativamente superior (P<0.01 y
P=0.02), respectivamente, que antes del tratamiento. En invierno,
los niveles de CK fueron menores (P<0.01) que en primavera. Se
concluye que, en general, el transporte asociado al ayuno produjo
mayores aumentos de las concentraciones sanguíneas de
cortisol, VGA, leucocitos y glucosa, siendo más estresante
que el confinamiento en ayuno.
SUMMARY
The aim of this study was to determine in steers the
effect of two periods of fasting, either confined on the farm or
transported by road, on their blood concentration of cortisol,
glucose, ß-HBA, lactate and the blood values of leucocytes;
PCV and plasmatic activity of CK. The study included two
experiments, carried out during July 2001 (winter) and November
2002 (spring), in the province of Valdivia, Chile.
One hundred and twenty steers from the same farm, breed,
age (milk teeth or 2 permanent teeth) and live weight (average
450 kg) were used. In each experiment (winter – spring), 60
steers were bled by coccigeous venopunction, identified
individually with ear tags and weighed. They were randomly
allocated into four groups of 15 steers each: one group was
transported by road for three hours without water or food, at the
same time and under similar conditions, another group was kept
confined in a pen at the farm. The other 2 groups were subjected
to respectively similar conditions, but for a period of 16 hours.
After treatments the steers were bled and weighed
again.
Plasma concentrations of cortisol were determined by
radioimmunoassay (RIA); the glucose concentrations by the GOD-PAP
test without deprotenization (GL 2623, RANDOX®), lactate
concentrations by the UVenzimatic method; ß-HBA by using
the enzymatic technique that uses the ß-hidroxibutirate
deshidrogenase enzyme for measuring the pass from NAD+ to NADH;
PCV and leucocytes values by using the SYSMEX KX – 4N
haematological counter and the CK plasma activity was measured by
the UV kinetic method. The stress indicators
were checked for a normal distribution by visual inspection of a
histogram of the variables with the Kolmogorov-Smirnov test. When
the distribution was not normal, the variables were normalised by
taking a natural logarithm. Multivariable generalised linear
regression models were used to determine the associations between
the stress indicators
and the independent variables. The independent variables were
treatment (before transport or confinement, fasting in
confinement for 3 hours, transported for 3 hours, fasting in
confinement for 16 hours, and transported for 16 hours), and
season (winter or spring).
Steers that were transported for 3 h and those fasted in
confinement for 16 h had significantly higher (P<0.01)
cortisol concentrations than before treatment; the other
treatments and season did not significantly (P=0.20) influence
the concentration of cortisol. PCV values were significantly
(P<0.01) decreased in steers confined for 3 h and
significantly (P=0.02; P<0.01) increased in the steers
transported for 3 and 16 h, respectively. The PCV values were
also higher (P<0.01)in winter than in spring. Leukocyte values
in steers did not show a consistent pattern but increased
(P<0.01) in steers that were transported both for 3 and 16 h.
Season did not influence leukocytes values. Glucose
concentrations significantly increased (P<0.01) with all
treatments. In winter, glucose was significantly (P<0.01)
lower than in spring. Steers that fasted in confinement for 3 h
had significantly lower (P=0.02) lactate concentrations than
before treatment. Steers in confinement for 16 h and those
transported for 16 h had significantly (P<0.01) lower
ß-HBA concentrations than before treatment. ß-HBA
concentrations were significantly (P<0.01) higher in winter.
Steers that were confined for 3 h and those transported for 3 h
had significantly (P<0.01 P= 0.02) higher CK levels than
before treatment. In winter, the CK activity was significantly
(P<0.01) lower than in spring.
